Publication: Sıçan (Wistar Albino) Merkezi Sinir Sisteminde Kadmiyum Sülfatla (Cdso4) Birlikte Verilen Nitrik Oksit Sentaz (Nos) İnhibitörlerinin Toplam Lipid ve Protein Miktarlarına Etkisi
Abstract
ÖZETSıçan (Wistar albino) Merkezi Sinir Sisteminde Kadmiyum Sülfatla (CdSO4)Birlikte Verilen Nitrik Oksit Sentaz (NOS) nhibitörlerininToplam Lipid ve Protein Miktarlarına EtkisiMehmet Emin ÖNGEROndokuz Mayıs Üniversitesi, Ocak 2007Sıçanlarda (Wistar albino) somatomotor kortekse kadmiyum sülfat (CdSO4)verildikten sonra spesifik Nitrik Oksit Sentaz (iNOS) enzim inhibitörü olanAminoguanidin (AG) ve NO öncüsü olan L-Arjinin (L-Arg)'in beyin ve beyinciktekitoplam lipid ve protein miktarlarına etkisini tespit etmek üzere, sunulan çalışmaplanlandı. Bu amaçla, saf, kontrol, kadmiyum+L-Arg ve kadmiyum+AG olmak üzere7'şer sıçandan oluşan 5 grup oluşturuldu. Kadmiyum grubu sıçanlara 0,0021 mg/kgCdSO4 sol hemisfere (bregmanın lateraline) intrakortikal yoldan enjekte edildi. Kontrolgrubuna aynı hacimde serum fizyolojik verildi. Kadmiyum+AG grubuna günde iki kezolmak üzere 100 mg/kg AG 15 gün süreyle intraperitoneal yoldan verildi.Kadmiyum+L-Arjinin grubuna da günde iki kez olmak üzere 1000 mg/kg L-Arjinin 15gün süreyle intraperitoneal yoldan verildi. Saf grup ise hiçbir işleme tabi tutulmadı vehiçbir madde verilmedi.Elde edilen sonuçlar, çalışılan diğer gruplara göre kadmiyumun toplam lipid veprotein miktarlarını düşürdüğünü ve toksik etkiye sahip olduğunu göstermektedir.Ayrıca, sunulan çalışma ile sıçanda kadmiyumun bu nörotoksik etkisinin, spesifik NOSinhibitörü olan AG tarafından azaltıldığına dair sonuçlar yeni bulgulardır. L-Arjinininkadmiyumdan kaynaklanan toplam lipid ve protein miktarlarındaki azalmayı arttırdığınıve bu nörotoksik etkide NO'nun da rol aldığını, elde ettiğimiz bulgular göstermektedir.ANAHTAR SÖZCÜKLER1. For this purpose 5 groups of rats, each containing 7animals were formed as non-operated, control, cadmium, Cd+L-Arg and Cd+AGrespectively. Rats in cadmium groups received 0,0021 mg/kg intracortical Cd injectionin the left hemisphere, lateral to Bregma. Control group received the same volume ofsaline. Cd+AG group received i.p AG injections for 15 days after the intracortical Cdinjections twice a day in 100 mg/kg doses, respectively. Cd+L-Arg group received i.pL-Arg injections for 15 days after the intracortical Cd injections twice a day in 1000mg/kg doses, respectively, too.Non-operated group received neither any treatment norany operation.In conclusion, it was determined that CdSO4 lowers the total lipid and proteinquantities, and affects the rats toxically when compared to other groups. In addition, ourresults are new findings suggesting that the toxic effect of CdSO4 was decreased byspesific NOS inhibitor AG in rat.
