Abstract
Bu çalışma, Nisan 2015 – Ekim 2017 tarihleri arasında Türkiye'nin Karadeniz kıyıları boyunca Padina, Adanson cinsine ait tür veya türlerin klasik ve moleküler yöntemler kullanılarak biyolojik çeşitliliğinin belirlenmesi amacı ile gerçekleştirilmiştir. Bu amaçla Karadeniz kıyısı boyunca 24 istasyondan örneklemeler yapılmıştır. Bunlardan 7 istasyonda Padina örnekleri bulunmuştur. Elde edilen örneklerin rbcL ve cox3 baz dizileri belirlenmiş ve bu diziler üzerinden analizler yürütülmüştür. Elde edilen dizilere uygulanan filogenetik analizler, morfolojik ve anatomik incelemeler sonucu örnekler P. pavonica olarak teşhis edilmiştir. Bu tez çalışması Karadeniz'de Padina cinsinin moleküler olarak teşhisine yönelik ilk araştırma olduğundan, bölgeden bu türe ait ilk moleküler kayıtları içermektedir. Yapılan bu çalışma sonucunda, Padina cinsinin Karadeniz'de P. pavonica ile temsil edildiği ve türün morfolojik olarak bir farklılaşma göstermemekle birlikte Akdeniz soy hattından ayrılmaya başladığı görülmüştür. Ayrıca son 20 yılda P. pavonica'nın Karadeniz'de dağılım gösterdiği alanların azaldığı tespit edilmiştir.
In this study, we aimed to determine species of the genus Padina along Black Sea coast of Turkey, and the biological diversity using conventional and molecular methods. For this purpose, between April 2015 and October 2017 samples were collected from 24 stations along the Black Sea coast. İn seven of these stations were found examples of genus Padina. The rbcL and cox3 base sequences of the obtained samples were determined and analyses were performed on these sequences. The phylogenetic, morphological and anatomical analyses of the obtained samples indicated that these samples were belong to the Padina pavonica. This thesis is the first study on the molecular diagnosis of the genus Padina in the Black Sea, and it contains the first molecular records of this species in the region. As a result of this study, it is observed that the genus Padina is represented by P. pavonica in the Black Sea and the species have begun to separate from the Mediterranean lineage, while showing no morphological differentiation. It has also been found that, the area of spread P. pavonica in the Black Sea during the last 20 years has decreased.
