Abstract
Bu çalışmada, 9 generasyon kendilenmiş 12 mısır genotipi, 7 local mısır genotipi ve 1 köylü genotipinde benzerlik ve ayırım dereceleri renk ölçüm cihazı ve moleküler markör kullanılarak değerlendirilmiştir. Bu çalışma kendilenmiş mısır hatlarının kalitesini etkileyen saflığın, daha ucuz ve hızlı yöntemle tespit edilmesi amacıyla hazırlanmıştır. Bu amaçla tohumda genetik safiyet, spektrofotometrik yöntem kullanılarak üç boyutlu olarak analiz edilmiştir. Bu yöntem, mısır ıslah programlarında ucuz ve kısa sürede yapılabilmesiyle alternatif yöntem olarak ele alınmıştır. Genotiplere ait renk ölçümü; embryo, sırt ve dik olmak üzere üç kısımdan alınmıştır. Bu ölçümlerden elde edilen L*, a* ve b* değerlerinden kümeleme, PCA ve PCoA analizleri ile genotiplerin birbirinden ayrıldığı belirlenmiştir. Genotipler üzerinde yapılan moleküler analizlerle de tam ayrım yapılmış ve renk analiz sonuçlarını desteklediği belirlenmiştir.
In this study, the genotypes similarity and distintion levels are analyzed among twelve maize genotypes selfed since nine generations, seven local maize and one villager maize genotypes using color spectrophotometer and molecular marker. This study was prepared so that the genetic purity affecting the quality of selfed maize lines is determated using the fastest and chapest methods. For this purpose, the genetic purity of seed were analyzed with three dimensions spectrophotometric method. This method is targeted as an alternative method to the other expensive and time consuming methods which establish the genetic purity in inbreding of the maize seed lines. Color measurement belong to these genotypes were taken from the top, back and embryo of seeds at three sides. The L*, a* and b* values were compared for each genotypes with/within each others using cluster analysis, PCA and PCoA analysis, statistically. It was determinated that these values can distinguish the genotypes. The molecular analysis made the complete separation among genotype and it was determined that molecular analysis supports the results of color analysis.
