dc.description.abstract | Bacteria and archaea in frozen (-20 degrees C) ruminal digesta were analysed by qPCR and cloning/sequencing of 16S rRNA genes. Samples frozen with and without glycerol as cryoprotectant indicated a major loss of Bacteroidetes in unprotected samples, resulting in higher proportions of Firmicutes. Archaeal numbers and diversity were unaffected. (C) 2013 Elsevier B.V. All rights reserved. | en_US |

dc.description.sponsorship | The Rowett Institute of Nutrition and Health is funded by the Rural and Environment Science and Analytical Services Division (RESAS) of the Scottish Government. BG thanks the Turkish Government for the award of a travelling fellowship. We thank Bob Mayes and Dave Hamilton of the James Hutton Institute for their permission and help in sampling the sheep digesta. Petra Louis and Freda McIntosh were generous with their invaluable advice on qPCR. Gillian Campbell and Pauline Young provided an excellent DNA sequencing service. | en_US |
