İki Önemli Baz Analoğu Ribavirin ve 5-Fluoruracil'in İnfeksiyöz Pankreatik Nekrozis (ıpN) Virusu Üzerinde Antiviral Etkisinin Araştırılması

Abstract

Bu araştırma projesi TÜBİTAK tarafından 1002 Hızlı Destek Programı kapsamında desteklenmiştir. Projede 2 farklı mutajenin; Ribavirin (RBV) ve 5-Fluorouracil (5-FU) İnfeksiyöz pankreatik nekrozis virus (IPNV) üzerine antiviral etkisi ve mutasyon oluşturma potansiyelleri, duyarlı hücre hattı EPC (Epithelioma Papulosum Cyprini) üzerinde in vitro olarak araştırılması hedeflenmiştir. Bu amaçla; ilk olarak RBV ve 5-FU sitotoksik dozları araştırılarak, sırası ile 50 µM/mL ve 100 µM/mL olarak tespit edilmiştir. Daha sonra IPNV'nin ilaç baskısı altında ve ilaç baskısı altında olmadan EPC hücre hattında seri pasajları gerçekleştirilmiştir. Her pasaj sonunda Real Time Revers -Transkriptaz Polimeraz Zincir Reaksiyonu ile konfirmasyonları gerçekleştirilmiştir. Yapılan seri pasajlar viral yük açısından Kantitatif Real Time PCR (qrt-PCR) ile kontrol edilmiştir ve viral yüklerde ilaç baskısı altında olmayan virus ile karşılaştırdığında viral yükte azalma açık olarak tespit edilmiştir. Ribavirin baskısı altında olan virusun 1.pasaj için 1,18x105 kopya/µl olan viral kopya sayısı, 5.pasaj sonunda 1,49x102 kopya/µl'e düşmüştür. 5-FU baskısı altında olan virusun, 1.pasaj için 3,84x103 kopya/µl olan viral kopya sayısı hemen her pasajda kademeli olarak azalarak (2.pasajda viral yük artmıştır) 5.pasaj sonunda 1,78x102 kopya/µl'e düşmüştür. Ribavirin ve 5-FU baskısı baskısı altında olan virusun ve ilaç baskısı altında olmayan virusun 1 ve 5.pasajları parsiyel kısmi sekans ile kontrol edilmiş ve bu mutajenlerin antiviral etkileri olmasına rağmen virusun VP2 üzerinde herhangi bir mutasyona neden olmadığı tespit edilmiştir. Sonuç olarak yaptığımız bu çalışmada iki mutajenin antiviral etkinliği yönünde veriler elde edilmilştir. Çalışma için seçilen gen bölgesinde herhangi bir mutasyon tespit edilmemesi nedeni ile diğer viral proteinler üzerine etkisi ve/veya virusun replikasyon siklusu üzerine etkisi araştırılması önerilir.

In the project, it was aimed to investigate the antiviral effect of 2 different mutagens; Ribavirin (RBV) and 5-Fluorouracil (5-FU) on Infectious Pancreatic Necrosis Virus (IPNV) and their mutation-inducing potential on the susceptible cell line EPC (Epithelioma Papulosum Cyprini) in vitro. For this purpose; firstly, cytotoxic doses of RBV and 5-FU were investigated and determined as 50 µM/mL and 100 µM/mL, respectively. Then, serial passages of IPNV were performed under and without drug suppression. At the end of each passage, confirmation was performed by Real Time rt-PCR. Serial passages were checked for viral load by qrt-PCR and a clear decrease in viral load was detected in viral loads compared to the virus not under drug suppression. The viral copy number of the ribavirin-suppressed virus decreased from 1.18x105 copies/µl for the 1st passage to 1.49x102 copies/µl at the end of the 5th passage, The viral copy number of the virus under 5-FU suppression decreased gradually with each passage from (2nd passage viral load increased) 3.84x103 copies/µl for the 1st passage to 1.78x102 copies/µl at the end of the 5th passage. Passages 1 and 5 of ribavirin-and 5-FU-suppressed virus and non-drug-suppressed virus were checked by partial partial sequence and it was found that although these mutagens had antiviral effects, they did not cause any mutation on VP2. In conclusion, in this study, we obtained data on the antiviral activity of two mutagens. Since no mutation was detected in the gene region selected for the study, it is recommended to investigate the effect on other viral proteins and/or the effect on the replication cycle of the virus.
