Abstract
Amaç: Çalışmamızda RASSF1A ve RASSF2A tümör baskılayıcı genlerinin hipermetilasyon durumlarının endometriyal kansere etkisinin incelenmesi amaçlanmıştır. Materyal Metod: Ondokuz Mayıs Üniversitesi Kadın Hastalıkları ve Doğum Kliniği' nde Endometriyal CA tanısı konan 15 hasta çalışmaya dâhil edildi. EDTA' lı tüplere periferik kan örnekleri alındı. Ayrıca, Endometriyal CA tanısı konulmuş bu 15 hastanın Ondokuz Mayıs Üniversitesi Patoloji bölümünden, formaline fikse edilmiş parafin gömülü tümör doku örnekleri temin edildi. Kontrol grubu için, Ondokuz Mayıs Üniversitesi Kadın Hastalıkları ve Doğum Kliniği'nden sezeryan doğum sırasında 10 sağlıklı kadından rahim doku örnekleri alındı. Periferik kan, rahim doku ve parafine gömülü tümör doku örneklerinden DNA izolasyonu yapıldı. Daha sonra DNA örneklerine bisülfit modifikasyon yöntemi uygulandı ve metilasyon spesifik PCR yapıldı. PCR ürünleri agaroz jelde yürütülerek, RASSF1A ve RASSF2A genlerinin promoter CpG metilasyon analizi yapıldı. Elde edilen sonuçlar SPSS ve Ki-kare analizi ile değerlendirildi. Bulgular: RASSF1A geni promotor bölgesi metilasyona spesifik PCR analizi sonuçları, hasta kanı, tümör dokusu ve kontrol grubu örnekleri ile karşılaştırıldığında istatistiksel olarak anlamlı bir fark saptanmazken (p=0,08), RASSF2A geni promotor bölgesi metilasyona spesifik PCR analizi sonuçları, hasta kanı, tümör dokusu ve kontrol grubu örnekleri ile karşılaştırıldığında istatistiksel olarak anlamlı derecede fark görüldü (p<0,001). Sonuç: Çalışmamızın sonucuna göre, Türk popülasyonunda RASSF1A geni promotor bölgesi metilasyonu endometrium kanseri gelişiminde önemli bir rol oynamazken, RASSF2A geni promotor bölgesi metilasyonunun önemli derecede rol oynadığı saptanmıştır. Aim: In this study, it is aimed to evaluate effects of RASSF1A and RASSF2A tumor suppressor genes hypermethylation status in endometrial cancer. Material and Method: We evaluated 15 endometrial CA diagnosed patients in Ondokuz Mayıs University Obstetrics and Gynecology Clinic. Peripheral blood samples were taken into EDTA tubes. Besides, formalin fixed paraffin embedded tumor tissue samples of these 15 patients were collected from Ondokuz Mayıs University Pathology Department. For control group, womb tissue samples of 10 healthy women were taken in Cesarian section delivery operation from Ondokuz Mayıs University Obstetrics and Gynecology Clinic. DNA isolations were performed from peripheral blood, womb tissue and paraffin embedded tumor tissue samples. Then, bisulfite modification method was applied to DNA samples and methylation specific PCR was performed. PCR products were analyzed in agarose gel and RASSF1A and RASSF2A genes' promoter CpG methylation status were registered. The results were evaluated with SPSS and Chi square statistical analysis. Results: As a result of the study, there is no statistical significant difference of RASSF1A gene promoter region methylation status between patients' blood and tumor tissue samples and healthy womb tissue samples (p=0.08). Also according to results of RASSF2A gene promoter region methylation status, there is statistical significant difference when comparing patients' blood and tissue samples and healthy womb tissue samples (p<0.001). Conclusion: According to results of our study, RASSF1A gene promoter region methylation status doesn't have an important role in endometrium cancer cases in Turkish population. However, RASSF2A gene promoter region methylation status has an important role in endometrium cancer cases in Turkish population.
