dc.description.abstractKeçiboynuzu gamı (LBG), biyoaktivite ve biyoteknolojik özellikleri olan eter ve hidroksil grupları gibi fonksiyonel gruplara sahip hidrofilik ve çok yönlü bir polisakkarittir. Nanoparçacıklar önemlidir, son yıllarda yaygın uygulama alanlarına sahiptir ve antibakteriyel aktiviteleri nedeniyle tıbbi alanlarda yaygın olarak kullanılmaktadır. Birçok araştırmacı nanopartikül sentezinde düşük maliyeti nedeniyle yeşil sentez yöntemini tercih etmektedir. Bu çalışmada gümüş nanopartikül (AgNP) yüklü keçiboynuzu gamı (LBG) / polivinil alkol (PVA) hidrojelleri, çözelti döküm yöntemi ile hazırlanmıştır. İncir yaprağı ekstresi (İYE) kullanılarak sentezlenen AgNP'ler UV-görünür spektroskopi ve taramalı elektron mikroskop (SEM) kullanılarak karakterize edildi. Geliştirilen gümüş nanoparçacık katkılı keçiboynuzu gamı - polivinil alkol hidrojellerinin kimyasal yapılarının tanımlanması ve hidrojellerin morfolojik karakterizasyonu için Fourier Transform-kızılötesi (FTIR) spektroskopisi, X ışını kırınımı (XRD) analizi ve Taramalı Elektron Mikroskobu (SEM) kullanıldı. Ayrıca, hidrojelin şişme davranışı farklı pH koşulları altında incelendi ve hidrojelerin termal stabilitesini incelemek için Termogravimetrik Analiz (TGA) kullanıldı. İncir yaprağı ekstresi (İYE) kullanılarak farklı AgNO3 konsantrasyonlarında sentezlenen AgNP'lerin antibakteriyel etkinliği, disk difüzyon yöntemi ile değerlendirildi. LBG/PVA/AgNP hidrojellerinin E. coli, P. aeruginosa gibi gram negatif bakterilere ve S. aureus, E. faecalis gibi gram pozitif bakterilere karşı antibakteriyel özellikleri Minimum İnhibisyon Konsantrasyon (MİK) yöntemi ve Disk difüzyon yöntemi kullanılarak değerlendirildi. İncir yaprağı ekstresi (İYE) kullanılarak sentezlenen AgNP'ler ve LBG/PVA/AgNP hidrojelleri, gram-negatif ve gram-pozitif bakteriye karşı iyi antibakteriyel aktivite gösterdiği gözlendi. Özetle, yeni hazırlanan LBG/PVA/AgNP hidrojelleri umut verici antibakteriyel özelliklere sahiptir.
dc.description.abstractLocust bean gum (LBG) is a hydrophilic and versatile polysaccharide possessing functional groups such as ether and hydroxyl groups which are areas useful for bioactivity and biotechnological field. Nanoparticles are important and nowadays they are widely used in medical fields due to their antibacterial activity. Many researchers prefer green route production and low cost of nanoparticle synthesis. In this work, silver nanoparticles (AgNPs) loaded locust bean gum (LBG)/ polyvinyl alcohol (PVA) hydrogels were prepared by the gel casting method. AgNPs synthesized by fig leaf extract (FLE) were characterized using UV-visible spectroscopy and scanning electron microscopy (SEM) in order to confirm the formation of AgNPs. The developed silver nanoparticle impregnated locust bean gum-polyvinyl alcohol hydrogels were characterized using Fourier Transform-Infrared (FTIR) spectroscopy, X-ray diffraction (XRD) analysis and Scanning Electron Microscopy (SEM) for identifying chemical structure such as chemical bonds and for the morphological characterizations of hydrogels, respectively. Additionally, the swelling behavior of the hydrogel was studied under different pH conditions and Thermogravimetric Analysis (TGA) was conducted to study the stability of hydrogel. The antibacterial activities of AgNPs synthesized using fig leaf extract (FLE) with different concentrations of AgNO3 were evaluated by the disc diffusion method. The antibacterial properties of LBG/PVA/AgNP hydrogels against gram-negative bacteria such as E. coli, P. aeruginosa and gram-positive bacteria such as S. aureus, E. faecalis were evaluated using the Minimum Inhibitory Concentration (MIC) method and Disc diffusion method. Finally, AgNPs synthesized using the fig leaf extract (FLE) and LBG/PVA/AgNP hydrogels showed good antibacterial activity against gram-negative and gram-positive bacteria. In summary, the newly prepared LBG/PVA/AgNP hydrogels have promising antibacterial properties.en_US
