dc.description.abstractV ABSTRACT RADIOGRAPHIC, BIOCHEMICAL, HISTOPATHOLOGIC, ULTRASTRUCTURAL EVALUATION IN LIGATURE-INDUCED PERIODONTITIS AND EXPERIMENTAL LATHYRISM MODEL Gonca ÇAYIR KELEŞ, Ph.D. Thesis University of Ondokuz Mayıs Samsun, September 2001 Lathyrism is characterized by defective collagen synthesis due to inhibition of lysyl oxidase which is essential for interfibrillar cross-linking. Lathyritic agent beta-aminoproprionitrile (P-APN) accepted as a suitable agent for the study of connective tissue metabolism. The concept of creating acute and chronic bone defects with ligatures has provided the strongest acceptance to obtain periodontitis like lesions in animals. Our main purpose is to compare ligature-induced periodontitis with experimental lathyrism in the manner of radiographic, biochemical, histopathological, and ultrastructural methods. In our study 45 Wistar rats weighting between 150-250gm were used. Ligature-induced periodontitis was created in 15 rats' molar teeth. Experimental lathyrism was obtained with daily subcutaneous injection of p-APN for 40 days in 15 rats. Fifteen rats were used as the control. At day 40, blood was taken by cardiac puncture, then all rats were decapitated. Activity of serum alkaline phosphatase were determined by optimum standart method. Left and right mandibles were dissected out; radiographs were taken, extraction test was performed on the right mandibular first molar. Histopathologic and ultrastructural evaluation was performed on the left mandibular molar teeth. Gingival biopsies taken from the same region were homogenized and IL-ip levels was assessed using the ELISA method. We observed the similar findings both in ligature-induced periodontitis and lathyritic group, unlike the control ones. Early results of our study suggest that the lathyritic rats might be used as an alternative model for experimental periodontitis.en_US
